RESUMO

Litchi (Litchi chinensis Sonn.) is a valuable subtropical fruit tree with high-quality fruit. However, its economic benefits and sustainable development are restrained by a number of challenges. One major challenge is the lack of extremely early and late maturing high-quality varieties due to limited availability of varieties suitable for commercial cultivation and outdated breeding methods, resulting in an imbalanced supply and low price of litchi. Flowering time is a crucial genetic factor influencing the maturation period of litchi. Our previous research has highlighted the pivotal role of the LcFT1 gene in regulating the flowering time of litchi and identified a gene associated with LcFT1 (named as LcSOC1) based on RNA-Seq and weight gene co-expression network (WGCNA) analysis. This study further investigated the function of LcSOC1. Subcellular localization analysis revealed that LcSOC1 is primarily localized in the nucleus, where it acts as a transcription factor. LcSOC1 overexpression in Nicotiana tabacum and Arabidopsis thaliana resulted in significant early flowering. Furthermore, LcSOC1 was found to be expressed in various tissues, with the highest expression in mature leaves. Analysis of spatial and temporal expression patterns of LcSOC1 in litchi varieties with different flowering time under low temperature treatment and across an annual cycle demonstrated that LcSOC1 is responsive to low temperature induction. Interestingly, early maturing varieties exhibited higher sensitivity to low temperature, with significantly premature induction of LcSOC1 expression relative to late maturing varieties. Activation of LcSOC1 triggered the transition of litchi into the flowering phase. These findings demonstrate that LcSOC1 plays a pivotal role in regulating the flowering process and determining the flowering time in litchi. Overall, this study provides theoretical guidance and important target genes for molecular breeding to regulate litchi production period.

Panum's limiting case is a phenomenon of monocular occlusion in binocular vision. This occurs when one object is occluded by the other object for one eye, but the two objects are both visible for the other eye. Although previous studies have found that vertical gradient of horizontal disparity and cue conflict are two important factors for double fusion, the effect of training on the sensitivity and stability of Panum's limiting case remains unknown. The current study trained 26 participants for 5 days with several of Panum's configurations (Gilliam, Frisby, and Wang series). The latency and duration of double fusion were recorded to examine the effects of training on sensitivity and stability of double fusion in Panum's limiting case. For each level of vertical gradient of horizontal disparity and cue conflict, the latency of double fusion decreased and the duration of double fusion increased with each additional training session. The results showed that vertical gradient of horizontal disparity and cue conflict interacted, and the duration of high cue conflict was significantly shorter than that of medium and low cue conflict for each level of vertical gradient of horizontal disparity. The findings suggest that there is an effect of training for vertical gradient of horizontal disparity and cue conflict in Panum's limiting case, and that the three factors jointly affect the sensitivity and stability of double fusion.

Vascular and neurological damage are the typical outcomes of ischemic strokes. Vascular endothelial cells (VECs), a substantial component of the blood-brain barrier (BBB), are necessary for normal cerebrovascular physiology. During an ischemic stroke (IS), changes in the brain endothelium can lead to a BBB rupture, inflammation, and vasogenic brain edema, and VECs are essential for neurotrophic effects and angiogenesis. Non-coding RNAs (nc-RNAs) are endogenous molecules, and brain ischemia quickly changes the expression patterns of several non-coding RNA types, such as microRNA (miRNA/miR), long non-coding RNA (lncRNA), and circular RNA (circRNA). Furthermore, vascular endothelium-associated nc-RNAs are important mediators in the maintenance of healthy cerebrovascular function. In order to better understand how VECs are regulated epigenetically during an IS, in this review, we attempted to assemble the molecular functions of nc-RNAs that are linked with VECs during an IS.

Aiming to look at the problems of the unreasonable layout of medical facilities and low coverage of primary medical services. This paper selects tertiary grade A hospitals, general hospitals, specialized hospitals, community-level hospitals, clinics, and pharmacies in the main urban areas of Chongqing as research objects. The nearest analysis, kernel density, mean center, and standard deviational ellipse method were used to analyze the spatial differentiation characteristics of medical facilities and public transportation stations. Spatial accessibility was assessed from the perspective of service area ratios and service population ratios by constructing multiple modes of transportation (pedestrian systems, bus lines, rail lines). The results show that (1) the spatial layout of medical facilities in the main urban area of Chongqing is unbalanced; and the spatial distribution of medical facilities is characterized by "large agglomeration, small dispersion" and "multi-center group"; (2) the sub-core circle is centered on the Southwest University Area in Beibei District, the University Town Area in Shapingba, the Yudong Area and Lijiatuo Area in Banan District, the Pingan Light Rail Station Area in Dadukou District, the Chongqing No. 8 Middle School Area in Jiulongpo District, the Tea Garden Area in Nanan District, and the Jiangbei Airport Area in Yubei District; (3) the medical facilities with the weakest average accessibility are tertiary grade A hospitals, and the strongest are pharmacies; (4) the areas with vital average accessibility are Yuzhong District, Shapingba District, Dadukou District, and Nanan District.

Objective: This study is aimed at exploring alteration in motor-related effective connectivity in individuals with transient ischemic attack (TIA). Methods: A total of 48 individuals with TIA and 41 age-matched and sex-matched healthy controls (HCs) were recruited for this study. The participants were scanned using MRI, and their clinical characteristics were collected. To investigate motor-related effective connectivity differences between individuals with TIA and HCs, the bilateral primary motor cortex (M1) was used as the regions of interest (ROIs) to perform a whole-brain Granger causality analysis (GCA). Furthermore, partial correlation was used to evaluate the relationship between GCA values and the clinical characteristics of individuals with TIA. Results: Compared with HCs, individuals with TIA demonstrated alterations in the effective connectivity between M1 and widely distributed brain regions involved in motor, visual, auditory, and sensory integration. In addition, GCA values were significantly correlated with high- and low-density lipoprotein cholesterols in individuals with TIA. Conclusion: This study provides important evidence for the alteration of motor-related effective connectivity in TIA, which reflects the abnormal information flow between different brain regions. This could help further elucidate the pathological mechanisms of motor impairment in individuals with TIA and provide a new perspective for future early diagnosis and intervention for TIA.

Social function impairment is the core deficit of autism spectrum disorder (ASD). Although many studies have investigated ASD through a variety of neuroimaging tools, its brain mechanism of social function remains unclear due to its complex and heterogeneous symptoms. The present study aimed to use resting-state functional magnetic imaging data to explore effective connectivity between the right temporoparietal junction (RTPJ), one of the key brain regions associated with social impairment of individuals with ASD, and the whole brain to further deepen our understanding of the neuropathological mechanism of ASD. This study involved 1,454 participants from 23 sites from the Autism Brain Imaging Data Exchange (ABIDE) public dataset, which included 618 individuals with ASD and 836 with typical development (TD). First, a voxel-wise Granger causality analysis (GCA) was conducted with the RTPJ selected as the region of interest (ROI) to investigate the differences in effective connectivity between the ASD and TD groups in every site. Next, to obtain further accurate and representative results, an image-based meta-analysis was implemented to further analyze the GCA results of each site. Our results demonstrated abnormal causal connectivity between the RTPJ and the widely distributed brain regions and that the connectivity has been associated with social impairment in individuals with ASD. The current study could help to further elucidate the pathological mechanisms of ASD and provides a new perspective for future research.

Cadmium (Cd) displays strong toxicity, high mobility, and cannot be degraded, which poses a serious threat to the environment. Cenococcum geophilum (C. geophilum) is one of the most common ectomycorrhizal fungi (ECMF) in the natural environment. In this study, three Cd sensitive and three Cd tolerant strains of C. geophilum were used to analyze the physiological and molecular responses to Cd exposure. The results showed that Cd inhibited the growth of all strains of C. geophilum but had a less toxic effect on the tolerant strains, which may be correlated to a lower content of Cd and higher activity of antioxidant enzymes in the mycelia of tolerant strains. Comparative transcriptomic analysis was used to identify differentially expressed genes (DEGs) of four selected C. geophilum strains after 2 mg/L Cd treatment. The results showed that the defense response of C. geophilum strain to Cd may be closely related to the differential expression of functional genes involved in cell membrane ion transport, macromolecular compound metabolism, and redox pathways. The results were further confirmed by RT-qPCR analysis. Collectively, this study provides useful information for elucidation of the Cd tolerance mechanism of ECMF.

BACKGROUND: Autism spectrum disorder (ASD) is associated with altered brain connectivity. Previous studies have focused on the static functional connectivity pattern from amygdala subregions in ASD while ignoring its dynamics. Considering that dynamic functional connectivity (dFC) can provide different perspectives, the present study aims to investigate the dFC pattern of the amygdala subregions in ASD patients. METHODS: Data of 618 ASD patients and 836 typical controls (TCs) of 30 sites were obtained from the Autism Brain Imaging Data Exchange (ABIDE) database. The sliding window approach was applied to conduct seed-based dFC analysis. The seed regions were bilateral basolateral (BLA) and centromedial-superficial amygdala (CSA). A two-sample t-test was done at each site. Image-based meta-analysis (IBMA) based on the results from all sites was performed. Correlation analysis was conducted between the dFC values and the clinical scores. RESULTS: The ASD patients showed lower dFC between the left BLA and the bilateral inferior temporal (ITG)/left superior frontal gyrus, between the right BLA and right ITG/right thalamus/left superior temporal gyrus, and between the right CSA and middle temporal gyrus. The ASD patients showed higher dFC between the left BLA and temporal lobe/right supramarginal gyrus, between the right BLA and left calcarine gyrus, and between the left CSA and left calcarine gyrus. Correlation analysis revealed that the symptom severity was positively correlated with the dFC between the bilateral BLA and ITG in ASD. CONCLUSIONS: Abnormal dFC of the specific amygdala subregions may provide new insights into the pathological mechanisms of ASD.

Introduction: Panum's limiting case is one of the typical configurations of monocular occlusion region. The matching rule of Panum's limiting case is the key to understanding how monocular occlusion region produces stereopsis. There are currently two main views on the matching rule of Panum's limiting case, namely double fusion and uniqueness constraint. This paper further discusses its matching mechanism on the basis of previous studies. Methods: In this study, fold line Panum's stimuli were used to study the matching rule of Panum's limiting case. In Experiment 1, fixation position was adopted to present the stimulus in a short time to explore the matching rules in Panum's limiting case. In Experiment 2, the effect of fixation position on Panum's limiting case matching results was further investigated. Results: The results of Experiment 1 show that when stimuli are presented in a short period of time, the reported result that a single feature in one eye may be matched alternately with two features in the other eye. This matching rule is called "fast alternative matching" in this article. The results of Experiment 2 results show that the position of the fixation could affect the matching result of participants. Conclusion: In conclusion, the matching rule of Panum's limiting case is fast alternative matching, and the matching result is related to the attention state of the participant. These results not only provide a new perspective for matching rules in Panum's limiting case, but also show that depth perception results in stereopsis can be influenced by top-down cognitive processing. This study provides a theoretical basis for studying the formation of stereopsis in the monocular region to a certain extent. In summary, the matching rule of Panum's limiting case is fast alternative matching. In previous studies, the perceived result of double fusion may be caused by fast alternative matching. Also, the matching results are related to the participant's state of attention, which suggests that the depth perception results of stereopsis are influenced by top-down cognitive processing.

MYB transcription factors have a wide range of functions in plant growth, hormone signaling, salt, and drought tolerances. In this study, two homologous transcription factors, PtrMYB55 and PtrMYB121, were isolated and their functions were elucidated. Tissue expression analysis revealed that PtrMYB55 and PtrMYB121 had a similar expression pattern, which had the highest expression in stems. Their expression continuously increased with the growth of poplar, and the expression of PtrMYB121 was significantly upregulated in the process. The full length of PtrMYB121 was 1395 bp, and encoded protein contained 464 amino acids including conserved R2 and R3 MYB domains. We overexpressed PtrMYB121 in Arabidopsis thaliana, and the transgenic lines had the wider xylem as compared with wild-type Arabidopsis. The contents of cellulose and lignin were obviously higher than those in wild-type materials, but there was no significant change in hemicellulose. Quantitative real-time PCR demonstrated that the key enzyme genes regulating the synthesis of lignin and cellulose were significantly upregulated in the transgenic lines. Furthermore, the effector-reporter experiment confirmed that PtrMYB121 bound directly to the promoters of genes relating to the synthesis of lignin and cellulose. These results suggest that PtrMYB121 may positively regulate the formation of secondary cell wall by promoting the synthesis of lignin and cellulose.

The present study investigates the effect of DPI resistance and inhalation flow rates on the lung deposition of orally inhaled mannitol dry powder. Mannitol powder radiolabeled with 99mTc-DTPA was inhaled from an Osmohaler™ by healthy human volunteers at 50-70L/min peak inhalation flow rate (PIFR) using both a low and high resistance Osmohaler™, and 110-130L/min PIFR using the low resistance Osmohaler™ (n=9). At 50-70L/min PIFR, the resistance of the Osmohaler™ did not significantly affect the total and peripheral lung deposition of inhaled mannitol [for low resistance Osmohaler™, 20% total lung deposition (TLD), 0.3 penetration index (PI); for high resistance Osmohaler™, 17% TLD, 0.23 PI]. Increasing the PIFR 50-70L/min to 110-130L/min (low resistance Osmohaler™) significantly reduced the total lung deposition (10% TLD) and the peripheral lung deposition (PI 0.21). The total lung deposition showed dependency on the in vitro FPF (R2=1.0). On the other hand, the PI had a stronger association with the MMAD (R2=1.0) than the FPF (R2=0.7). In conclusion the resistance of Osmohaler™ did not significantly affect the total and regional lung deposition at 50-70L/min PIFR. Instead, the total and regional lung depositions are dependent on the particle size of the aerosol and inhalation flow rate, the latter itself affecting the particle size distribution.
